海水中的致病菌包括來自生活污水、城市徑流以及海岸沙灘流入的陸源性致病菌及自然海水中致病菌。海水中致病菌的污染不僅威脅人們的健康安全，而且會(huì)對水產(chǎn)養(yǎng)殖業(yè)帶來巨大的危害和損失。因此，發(fā)展快速、準(zhǔn)確、高靈敏檢測海水中主要致病菌的傳感器技術(shù)受到了廣泛關(guān)注。 近期，中國科學(xué)院煙臺(tái)海岸帶研究所海洋環(huán)境電化學(xué)與傳感器技術(shù)研究團(tuán)隊(duì)和海岸帶微型生物生態(tài)與應(yīng)用團(tuán)隊(duì)合作，在前期研究工作的基礎(chǔ)(Analytical Chemistry, 2012, 84, 2055-2031)上，進(jìn)一步發(fā)展了一種免標(biāo)記、免固定化電位型傳感器用于海水致病菌的檢測。 該檢測系統(tǒng)以核酸適體作為識別分子，魚精蛋白作為指示離子，避免了對核酸適體的標(biāo)記和固定化，從而實(shí)現(xiàn)了對致病菌的快速、高效、低成本檢測。海水高鹽度能夠降低核酸適體和靶致病菌的結(jié)合能力，并會(huì)干擾聚離子選擇性電極的電位響應(yīng)。 在實(shí)際樣品分析中，研究采用了樣品在線快速過濾法，有效消除海水基體效應(yīng)，并通過致病菌富集提高了檢測靈敏度，滿足了實(shí)際海水檢測的要求。以單增李斯特菌為例，該檢測系統(tǒng)的檢出限達(dá)到了10 CFU mL-1。 該研究成果近期發(fā)表在國際化學(xué)刊物Analytical Chemistry 上。 原文摘要： Previous ArticleNext ArticleArticles ASAPPotentiometric Aptasensing of Listeria monocytogenes Using Protamine as an Indicator Jiawang Ding, Jiahong Lei, Xia Ma, Jun Gong and Wei Qin Exposure to pathogens in recreational or drinking water is a serious public health concern. It is important to rapidly determine and identify trace levels of pathogens in real environmental samples. We report here on a label-free potentiometric aptasensor for rapid, sensitive, and selective detection of Listeria monocytogenes (LM), a pathogen widely distributed in the environment. An aptamer binds specifically to internalin A, a surface protein present in LM cells. The target-binding event prevents the aptamer from electrostatically interacting with protamine, which can be sensitively detected using a polycation-sensitive membrane electrode. Using this method, LM can be detected down to 10 CFU mL–1. Coupled to an online filtration system, the bioassay has been evaluated with spiked coastal seawater samples and shows good recovery and high accuracy. This work demonstrates the possibility of developing potentiometric aptasensors for determination and identification of various bacteria in environmental samples.
